Objective:To investigate the role of TRIM56 across various tumors and validate its expression and function in esophageal cancer tissues and cells. Methods:The role of TRIM56 in different tumors was comprehensively analyzed through databases including UCSC, TCGA, GTEx, and GEPIA2, focusing on gene expression, prognostic significance, and immune cell infiltration. The expression of TRIM56 in Eca‑109 cells was assessed using qRT‑PCR and Western blot, while immunohistochemistry was employed to analyze TRIM56 levels in esophageal cancer tissues and adjacent non‑cancerous tissues. Transwell and scratch assays were conducted to evaluate the effects of TRIM56 overexpression and silencing on the invasive and migratory abilities of Eca‑109 cells. Results:TRIM56 expression was significantly elevated in most tumor tissues compared to normal tissues (P<0.05). High TRIM56 expression correlated with poor prognosis and increased immune cell infiltration in several tumor types. TRIM56 was found to be involved in critical biological processes such as innate immunity, tumor proliferation, and autophagy. Immunohistochemistry demonstrated that TRIM56 levels were significantly higher in esophageal squamous carcinoma tissues compared to adjacent non‑cancerous tissues. Functional assays revealed that overexpression of TRIM56 enhanced the invasive and migratory abilities of Eca‑109 cells, while silencing TRIM56 markedly inhibited these capabilities. Conclusion:TRIM56 expression is closely associated with clinical prognosis and immune cell infiltration in cancer patients. In esophageal cancer, high TRIM56 expression promotes cell invasion and migration, suggesting that TRIM56 functions as an oncogene. TRIM56 holds promise as a potential diagnostic and therapeutic target in esophageal cancer, offering new insights into its role in tumor progression.
